The structure of the macromolecular units on the cell walls of Bacillus polymyxa.

چکیده
Electron micrographs of negatively stained preparations of cell walls of Bacillus polymyxa have been investigated by optical diffraction and optical filtering techniques. Images of single layers of the cell wall, from which the ' noise' has been filtered optically, show hollow, squareshaped morphological units arranged on a square lattice of side 100 A. Single-layer images showing the same pattern have been filtered from moir6 patterns arising from two overlapping single layers. The morphological units are composed of four smaller subunits. The optical diffraction patterns from regions of two overlapping layers show extra reflexions which are attributed to multiple electron scattering.

Distribution of mannosamine and mannosaminuronic acid among cell walls of Bacillus species.
The distribution of mannosamine, mannosaminuronic acid, and the enzymes responsible for the formation of these saccharides was studied in nine species (18 strains) of Bacillus. Whereas UDP-N-acetylglucosamine 2-epimerase activity was detected in all of the strains examined, UDP-N-acetylmannosamine dehydrogenase, as well as the activity incorporating N-acetylmannosaminuronic acid residues from U...
متن کاملTransformation of Bacillus polymyxa with plasmid DNA.
A plasmid transformation system was developed for Bacillus polymyxa ATCC 12321 and derivatives of this strain. The method utilizes a penicillin-treated-cell technique to facilitate uptake of the plasmid DNA. Low-frequency transformation (10(-6) per recipient cell) of plasmids pC194, pBD64, and pBC16 was accomplished with this method. Selection for the transformants was accomplished on both hype...

Plant pathogenic microorganisms produce a variety of enzymes capable of degrading different polysaccharides of the plant cell walls. Pathogens use these enzymes to penetrate and colonize host cells. Polygalacturonases are thought to be the first cell wall-degrading enzymes secreted by pathogens when they grow on plant cell walls. Oligogalacturonic acids with the polymerization degrees of 10 to ...

متن کاملIn Silico Prediction of B-Cell and T-Cell Epitopes of Protective Antigen of Bacillus anthracis in Development of Vaccines Against Anthrax
Protective antigen (PA), a subunit of anthrax toxin from Bacillus anthracis, is known as a dominant component in subunit vaccines in protection against anthrax. In order to avoid the side effects of live attenuated and killed organisms, the use of linear neutralizing epitopes of PA is recommended in order to design recombinant vaccines. The present study is aimed at determining the dominant epi...
